SÓLIDOS SOLUBLES EN ZUMO DE UVA, ZUMO DE UVA RECONSTITUIDO, ZUMO DE UVA CONCENTRADO Y NÉCTAR DE UVA
Método de tipo I
LA ASAMBLEA GENERAL,
VISTO el artículo 2, párrafo iv del Acuerdo del 3 de abril de 2001 por el que se crea la Organización Internacional de la Viña y el Vino,
A PROPUESTA de la Subcomisión “Métodos de Análisis”,
CONSIDERANDO la propuesta de aplicar el método OIV-MA-AS2-02 del Compendio de Métodos Internacionales de Análisis de Vinos y Mostos al zumo de uva, el zumo de uva reconstituido, el zumo de uva concentrado y el néctar de uva,
DECIDE incorporar el siguiente método:
SÓLIDOS SOLUBLES EN ZUMO DE UVA, ZUMO DE UVA RECONSTITUIDO, ZUMO DE UVA CONCENTRADO Y NÉCTAR DE UVA
1. Ámbito de aplicación
El presente método permite determinar los sólidos solubles en el zumo de uva, el zumo de uva reconstituido, el zumo de uva concentrado y el néctar de uva en el intervalo de concentraciones de 10,0 °Bx a 69,2 °Bx.
2. Fundamento
El contenido de sólidos solubles de una muestra se determina a partir del índice de refracción a 20 °C, absoluto o en porcentaje en masa de sacarosa (grados Brix), que se lleva a la tabla II para obtener el contenido de azúcares en gramos por litro y en gramos por kilogramo de zumo de uva, zumo de uva reconstituido, zumo de uva concentrado o néctar de uva.
3. Equipo
3.1. Refractómetro de Abbe
El refractómetro debe estar provisto de una escala que indique:
- el porcentaje en masa de sacarosa con una aproximación del 0,1 %, o
- el índice de refracción con cuatro cifras decimales.
El refractómetro debe estar provisto de un termómetro cuya escala abarque por lo menos de 15 °C a 25 °C, así como de un dispositivo de circulación de agua que permita realizar las determinaciones a una temperatura de 20 °C ± 5 °C. Seguir estrictamente las instrucciones del instrumento, en especial en lo relativo a la calibración y al haz de luz.
Nota: También pueden utilizarse refractómetros electrónicos. En ese caso, no es necesario corregir el efecto de la temperatura si el equipo lo tiene en cuenta de forma automática.
3.2. Balanza analítica verificada y calibrada, con una precisión mínima de 0,01 g
4. Preparación de la muestra
Filtrar la muestra con un filtro de papel o una gasa seca doblada en cuatro; descartar las primeras gotas del filtrado y proceder a la determinación del producto filtrado.
En el caso del zumo de uva concentrado, dependiendo de su concentración, utilizar la muestra directamente o preparar una dilución pesando 200 g de zumo de uva concentrado y añadiendo agua purificada para uso en análisis de laboratorio (EN ISO 3696) hasta 500 g (F = 2,5).
5. Procedimiento
Llevar la muestra a una temperatura de unos 20 °C. Poner una pequeña cantidad en el prisma inferior del refractómetro, teniendo cuidado de que cubra uniformemente la superficie del vidrio, ya que los prismas están en contacto estrecho. Realizar la medición siguiendo las instrucciones del fabricante del instrumento. Leer el porcentaje en masa de sacarosa con una cifra decimal o el índice de refracción con cuatro cifras decimales. Efectuar por lo menos dos determinaciones de cada muestra preparada. Anotar la temperatura t en °C.
6. Cálculos
6.1. Corrección de temperatura
- Instrumentos graduados en porcentaje en masa de sacarosa: utilizar la tabla I para obtener la corrección de temperatura.
Tabla I. Corrección que debe aplicarse cuando el porcentaje en masa de sacarosa se determina a una temperatura distinta de 20 °C.

- Instrumentos graduados en índice de refracción: buscar el índice de refracción medido (columna 2) en la tabla II para obtener el valor correspondiente en porcentaje en masa de sacarosa (columna 1). Utilizar la tabla I para corregir el efecto de la temperatura y expresar el valor como concentración a 20 °C.

7. Expresión de los resultados
Los resultados se expresan en porcentaje en masa de sacarosa m/m (grados Brix), con dos cifras decimales y a partir de la media de dos determinaciones. En el caso del zumo de uva concentrado, multiplicar el resultado por el factor de dilución (F = 2,5).
8. Características del método (a título ilustrativo)
Se llevó a cabo un estudio de validación para evaluar la adecuación del método a la matriz zumo de uva. Se determinaron la linealidad, los límites de detección y cuantificación y la exactitud del método. La exactitud se determinó a partir de la precisión y la veracidad del método.
8.1. Linealidad del método
Según los resultados del análisis de regresión lineal, el método es lineal en el intervalo de 10,0 °Bx a 69,2 °Bx, como se indica en la tabla III.
8.2. Límite de detección y límite de cuantificación
El límite de detección (LD) y el límite de cuantificación (LC) se calcularon analizando un zumo de uva diluido. El LD es igual a 3 veces la desviación estándar y el LC es igual a 10 veces la desviación estándar (tabla III).
8.3. Precisión del método
Se tuvieron en cuenta la repetibilidad y la reproducibilidad, cuyos valores figuran en la tabla III. La repetibilidad se determinó como la desviación estándar de la repetibilidad (Sr) de 7 mediciones de las siguientes concentraciones: 15,9 °Bx y 69,2 °Bx.
La precisión intermedia (PI) se determinó como la desviación estándar (SPI) de mediciones con una misma muestra de zumo de uva realizadas por distintos operadores.
